No you need the dash (or its equivalent) if you want to keep that sentence structure. This is correct: "Which class should I choose between—Calligraphy or N5 Grammar?" If you really don't want to use the dash you can say "Should I choose Calligraphy or N5 Grammar? Both classes sound great!"
